چکیده

Increasingly, biochemical and genetic evidence indicates that mutations in the gene encoding p16, an inhibitor of cyclin-dependent kinases, may play a role in some forms of hereditary and sporadic tumors.

Inhibition of Breast Tumor Cell Growth by Ectopic Expression of p16/INK4A Via Combined Effects of Cell Cycle Arrest, Senescence and Apoptotic Induction, and Angiogenesis Inhibition

p16-mediated inhibition of cancer cell proliferation and tumor suppression have been studied before,; the common consensus is that p16's cell-cycle arrest function plays a primary role in these actions, with some additional apoptotic induction by p16. However, other effects of p16 that may potentially contribute to p16-mediated anti-tumor ability have not been well studied. The emerging data in...

The Molecular Balancing Act of p16 in Cancer and Aging

p16, located on chromosome 9p21.3, is lost among a cluster of neighboring tumor suppressor genes. Although it is classically known for its capacity to inhibit cyclin-dependent kinase (CDK) activity, p16 is not just a one-trick pony. Long-term p16 expression pushes cells to enter senescence, an irreversible cell-cycle arrest that precludes the growth of would-be cancer cells but also contributes...

Role of the Alternative INK4A Proteins in Human Keratinocyte Senescence: Evidence for the Specific Inactivation of p16 upon Immortalization

The INK4A locus on human chromosome 9p21 encodes two genes that have been implicated in replicative senescence and tumor suppression, p16 and p14. In contrast to p16, which is up-regulated to high levels, we were unable to detect p14 protein in senescent human keratinocytes. Also, p53, an established target of p14, did not increase, suggesting that p14 is not instrumental in human keratinocyte ...

A CDKN2A Mutation in Familial Melanoma that Abrogates Binding of p16 to CDK4 but not CDK6

The CDKN2A locus encodes two distinct proteins, p16 and p14, both of which are implicated in replicative senescence and tumor suppression in different contexts. Here, we describe the characterization of a novel strain of human diploid fibroblasts (designated Milan HDFs) from an individual who is homozygous for the R24P mutation in p16. As this mutation occurs in the first exon of INK4a (exon 1A...

Replacement of the p16/CDKN2 gene suppresses human glioma cell growth.

The p16/CDKN2 gene has many features of a growth suppressor gene: it maps to 9p21, a frequent region of loss of heterozygozity in a variety of tumor types; it encodes an inhibitor of cyclin-dependent kinase 4; and its homozygous deletion is common in tumor-derived cell lines. However, the lower frequency of alteration of the gene in primary tumor tissue as compared to the cognate tumor cell lin...
